A Month of Collaboration with Professors Richards and Symes: Scientific and Cultural Exchange at Theoreo
An intense and fruitful month of collaboration has just concluded at Theoreo, during which we had the honor of once again hosting Professors Sean M. Richards and Steven J.K. Symes from the University of Tennessee at Chattanooga (USA), seven years after their first visit to our laboratories.
The two academics chose to spend their sabbatical month in Italy, working in our laboratories on a series of innovative scientific projects that will soon be published in prestigious scientific journals. This period of intensive activity has further strengthened the collaboration between our team and these distinguished scientists.
During their stay, Professors Richards and Symes also had the opportunity to visit the laboratories of the Department of Medicine and Surgery at the University of Salerno and to deliver an outstanding seminar at the Department of Chemistry and Biology. The event provided a valuable opportunity for the local academic community to benefit from their expertise and scientific vision.
This month represented an extraordinary opportunity for cultural and scientific exchange for the entire Theoreo team. The dedication, rigor, and vision of Professors Richards and Symes left an indelible mark, further consolidating the long-standing and fruitful collaboration that has united us for years.
We express our deepest gratitude for the time and commitment that the two academics dedicated to this experience, and we are confident that our relationship will continue to flourish, paving the way for new opportunities in research and innovation.

OUR CEO @ TEDx Battipaglia
Our CEO, Jacopo Troisi, recently had the privilege of speaking at TEDx Battipaglia, where he delivered an insightful talk on emerging trends in medicine titled: “Towards the Medicine of the Future: Chronicles of a Journey.”
Over time, medicine has undergone several phases of transformation: from paternalistic medicine, where the doctor was the central figure, to evidence-based medicine, inspired by the pioneering research of Josef Dietl.
Today, we are witnessing a new and decisive transition toward personalized medicine, which, for the first time, shifts the focus from the disease to the individual in all their uniqueness.
Prof. Troisi delved into how metabolomics, the study of metabolites and biochemical processes, is becoming a cornerstone of this new paradigm.
Metabolomics not only provides new tools for more precise and timely diagnoses but also enables the development of personalized therapies, optimizing treatments based on the unique metabolic characteristics of each patient.
This represents a fundamental step toward a truly tailored medical model, with translational applications that promise to revolutionize and optimize patient management at every stage.

10th International Human Microbiome Consortium Congress
On the occasion of World Microbiome Day, it is essential to acknowledge the growing efforts of the international scientific community to investigate the implications of the microbiota and its metabolism on human health.
In recent days, six delegates from Theoreo attended the 10th International Human Microbiome Consortium Congress, held at the “Auditorium della Tecnica” in Rome. This event provided an important platform for scientific exchange, where our team members had the opportunity to discuss various projects in which Theoreo has played a key role in studying gut microbial metabolism.
Among the topics discussed were the preliminary findings from fecal metabolomic investigations within the GEMMA project (Genome, Environment, Microbiome, and Metabolome in Autism).
Increasing evidence in the literature suggests that microbial metabolites may play a pivotal role in various neurodevelopmental disorders, particularly through the gut-brain axis. Many research groups are currently investigating the mechanisms underlying this complex bidirectional communication system, and the GEMMA project is making a substantial contribution to this field in the context of autism spectrum disorders.
The Theoreo team is actively working to identify alterations in gut microbial metabolism associated with the condition.

OLIVIA DENTON
We bid a fond farewell to Olivia Denton, a graduate of the University of Tennessee at Chattanooga, who recently enriched the Theoreo team with her talent during a one-month internship. Olivia, a student of the renowned Prof. Sean Richards, joined us from the United States, demonstrating from the outset an extraordinary dedication and excellent research aptitude.
During her time with us, Olivia fully immersed herself in the challenges and opportunities offered by metabolomics, showcasing expertise from chemical sample analysis to data manipulation and the drafting of scientific articles destined for international journals. Her commitment and enthusiasm made her an invaluable asset to our team, while her joy and eagerness to learn helped foster a stimulating and collaborative work environment.
As Olivia embarks on her journey back to the United States to continue her studies in medicine, we are confident that her future is destined to shine. We look forward to collaborating with her again in the future and wish her the very best for all her new endeavors. Good luck, Olivia!

EVOLUTION OF CANCER PHARMACOLOGY
The Theoreo team recently took part in the "Evolution of Cancer Pharmacology" conference, a prestigious event organized by the Italian Society of Pharmacology (SIF) at the Grand Hotel Salerno on March 15th and 16th. During this engaging conference, participants delved into the revolution of cancer pharmacology, from preclinical research to clinical studies. The discussions highlighted the significant progress made in the treatment of oncological diseases over the past decades, thanks to the introduction of targeted therapies and immunotherapies. These advancements have been made possible through continuous research in the field of personalized medicine. The Theoreo team was thrilled to be part of this transformation and grateful for the opportunity to contribute to the ongoing evolution of research in this crucial area of medicine.
Among the contributions from Theoreo were three presentations: "A Metabolomic Approach Enhanced by Machine Learning for Colorectal Cancer Screening"; "Advancing Male Uro-genital Cancer Screening with Metabolomic Profiling and Ensemble Machine Learning"; and "Metabolomic Profiling for Non-Invasive Ovarian Cancer Screening." These works demonstrate the team's commitment to leveraging cutting-edge techniques, such as metabolomics and machine learning, to improve cancer screening and diagnosis, ultimately aiming to enhance patient outcomes in the fight against cancer.

Our CEO is currently in Granada, Spain, where he has been kindly invited to visit the prestigious Medina Foundation, a renowned non-profit research center born in collaboration with the University of Granada. BREAKING BARRIERS IN CANCER SCREENING
We are excited to share another great news from the Metabolomics Society conference. This year, our Chief Scientific Officer, Martina Lombardi, presented an extraordinary poster titled 'Breaking Barriers in Cancer Screening: Integrating Metabolomics Profiling and Ensemble Machine Learning for Comprehensive Gender-Specific Cancer Screening.'
Our goal with this project is to revolutionize cancer screening by developing an innovative system that, through the analysis of a single blood sample, can detect various types of tumors. In the specific case presented, we focused on male urogenital tract tumors, and the results were astonishing.
This research requires significant effort and a crucial component of artificial intelligence. Our researchers are tirelessly working to test the system on a broader cohort and further optimize it. Simultaneously, they are developing a similar test for the female urogenital tract.
The current landscape of cancer screening presents several challenges, including the low participation of the target population, often discouraged by the multiplicity of invasive tests. Our aim is to overcome these barriers by offering a single comprehensive, non-invasive test capable of detecting multiple types of tumors. Such a significant step forward could make a difference in many people's lives.
Research is at the heart of everything we do. We firmly support the importance of scientific research in improving health and well-being for all. We want to express our gratitude to Martina Lombardi and all the researchers tirelessly working on this groundbreaking project.
We will continue to invest resources and effort in research, with the goal of making significant progress in the fight against cancer. A better future for everyone's health is within our reach.

METABOLOMICS SOCIETY ANNUAL MEETING 2023
The time has finally come for the annual meeting with colleagues from around the world at the prestigious Metabolomics Society conference, held this year in Canada at the Niagara Falls Convention Centre.
Once again, Theoreo took center stage with an extraordinary presentation by Prof. Jacopo Troisi on the remarkable potential of Metabolomics profiling in screening for fetal malformations.
The evidence presented by Prof. Troisi is the result of a decade of in-depth studies involving approximately 3000 pregnant women, engaging researchers and hospitals both in Italy and New Zealand. These studies provide solid support for the use of metabolomics in screening for fetal malformations, and what's even more fascinating is that maternal biochemical response has been consistent regardless of the type of fetal malformation.
The presentation has sparked immense interest, and we are deeply proud of it. We firmly believe that supporting scientific research is crucial, as only through continuous progress can we improve our understanding of the mechanisms underlying human pathologies and offer new solutions to clinical practice.
We would like to express sincere thanks to Prof. Jacopo Troisi for his extraordinary commitment and dedication, as well as to all the researchers who have contributed to this important project. We will continue to work together to promote the well-being of mothers and children worldwide.

WELL-BEING AND PREVENTION DAY
A new appointment of the "Wellness Days" organized by Theoreo, this time at the Municipality of Battipaglia. We started the day with a brief informative seminar on female oncological prevention, led by Prof. Jacopo Troisi, CEO of Theoreo, Prof. Pierpaolo Cavallo, and Dr. Vera Abenante, Lab Manager of Theoreo. The seminar attracted a large audience and sparked great interest among the attendees.
Subsequently, we offered about 200 MEDEA tests for endometrial carcinoma screening free of charge to the women of the area who decided to participate in the study, entrusting themselves to scientific progress and becoming active participants.
We want to sincerely thank the municipal administration of the Municipality of Battipaglia, especially Mayor Dr. Cecilia Francese, for enthusiastically and sensitively welcoming our project.
A heartfelt thanks goes to the Inner Wheel of Battipaglia for their valuable logistical support and generous donation to research. Your dedication is an example for all of us. 💙

WELL-BEING AND PREVENTION DAY
After the great success of the last screening campaign, where Theoreo offered over 80 free prevention appointments, we are pleased to announce a new day dedicated entirely to women's health.
Our CEO, Prof. Jacopo Troisi, flanked by Prof. Pierpaolo Cavallo and Dr. Martina Lombardi, scientific head of Theoreo, will hold a short informative seminar on women's cancer prevention.
Following this, Theoreo staff will engage in the free administration of the MEDEA test for screening endometrial cancer, the most common cancer of the female reproductive tract in Western populations, for which, until now, no screening system had been available.
The test, which is rapid, painless and noninvasive, requires the drawing of a single drop of blood using a lancet, similar to measuring blood glucose.

FREE OPENDAY FOR WOMEN'S CANCER PREVENTION, AS PART OF THE CITY OF PONTECAGNANO FAIANO'S "OLTRE l'8 MARZO" EVENT
We are happy to announce that on 03/27/2023 at our office at 57 Lucania Street, Pontecagnano Faiano, there will be an open day with free examinations, entirely dedicated to women's prevention.
They will be carried out free of charge with prior reservation:
✓ Visit with Dr. Luigi Cremone, a breast surgeon. An appointment for a mammography examination, also free of charge, will also be provided for women who have received doubtful or unfavorable results from the breast evaluation, on a date to be arranged at the office of Diagnostica Cavallo of Drs. Armando and Pierpaolo Cavallo.
✓ Visit with Dr. Valentina Reina, a medical doctor specializing in Aesthetic Medicine, for the prevention and treatment of skin blemishes and skin damage caused by solar radiation.
✓ Nutritional counseling with Dr. Chiara D'Antonio, a nutrition biologist, in which an assessment of the patient's physical and nutritional status will be carried out with the aim of identifying any unhealthy eating habits and re-educating or rehabilitating the patient nutritionally and dietarily.
✓ MEDEA test, an innovative screening system for endometrial cancer, developed by Theoreo srl in collaboration with the G. Pascale Cancer Institute and the University of Salerno. The test consists of taking a single drop of blood by digital puncture. The sample will undergo metabolomic analysis in order to identify patients with the disease early. The above test is dedicated to women over the age of 50.

Metabolomics in Public Health: new screening methods for endometrial cancer
Our CEO had a talk at the GYNOCARE COST Action Conference, "Bridging the gap between Research and Cure in Rare Gynaecological Cancers: Where do we stand?" held Feb. 17 and 18 at the University of Campania "Luigi Vanvitelli" in Naples.
The conference was in collaboration with European Network for Gynaecological Rare Cancer research: From Concept to Cure.
The invited international speakers,shared insights on recent developments in gynecological rare cancers, focusing on:
- Basic and translational research: present and future
- Clinical trials: designing cures
- Difficult diagnosis: from the classical method to digital pathology
- Pharma-Biotech: the challenges of the 21st century
Theoreo's talk was titled "Development and Validation of a Serum Metabolomic Signature for Endometrial Cancer Screening in Postmenopausal Women."

Metabolomics in Public Health: new screening methods for endometrial cancer
On December 16, 2022, Theoreo in collaboration with prestigious research and treatment institutions will organize the first "Metabolomics in Public Health" conference. This first year has been dedicated to new methods for screening endometrial cancer.
Endometrial cancer represents the most common neoplasm of the female reproductive system, with more than 88,000 diagnoses per year in Europe. Although the prognosis is closely linked to early diagnosis, there is currently no mass screening for early detection of this cancer.
To effectively combat this disease, Theoreo and Hosmotic, two Campania-based companies engaged in scientific research in biotechnology, have joined efforts with authoritative Campania university and clinical institutions including the University of Salerno, the University of Naples Federico II, the IRCCS fondazione "G. Pascale" and the Avellino section of LILT (Lega Italiana per la Lotta ai Tumori) to create a tool that can be used as a secondary prevention tool, thus for population screening.
The free event was attended by numerous experts in the field and was accredited for CME credits for physicians, biologists and pharmacists, chemists and physicists.
At the end a benefit concert offered by AFI (Asoociazione Flautisti Italiani), the proceeds of which were donated to the assozioen ACTO Campania (Association Against Ovarian Cancer)

IV CONGRESS OF THE NORMAMERICAN SOCIETY OF METABOLOMICS (MANA)
The MANA (Metabolomics Association of North America) conference, was held Sept. 16-18.
Our CEO Jacopo Troisi (CEO) and Martina Lombardi (CSO) are located together with Prof. Sean Richards and Prof. Steven Symes (the former teaches Biology, the latter Chemistry at the University of Tennessee, Chattanooga) represented Theoreo at this important scientific event.
They presented the results of the latest validation of the Endometrial Carcinoma Screening Test (Medea Project), a noninvasive test to diagnose early this cancer that afflicts more and more women worldwide.
In addition, a poster on the GEMMA study, an international project in which we are involved in the analysis of autism spectrum disorders; one on data on the screening test for fetal malformations called "Mama Test"; one on our work about a unified approach to cancer screening by metabolomic profiling; and discussion on hepatic steatosis and its evolution to hepatocarcinoma.

MAMA TEST VALIDATION FIRST RESULTS PUBLISHED
It is now available online the paper summarizing the results of a long and complex validation trial of the MAMA Test. With support from the New Zealand arm of the SCOPE study led by Prof. Lesley McCowan of Auckalnd University, researchers at Theoreo blindly applied the MAMA Test to a collection of more than 2,000 pregnant women serum samples from New Zealand. The test correctly detected more than 99% of fetal malformations, confirming the MAMA Test as an accurate and noninvasive ally for early prenatal diagnosis.
The results were published in the prestigious journal AJOG - American Journal of Obstetrics and Gynecology. The papeer is freely accessible until 3rd Dec 2022 at this link.

18TH INTERNATIONAL CONFERENCE OF THE METABOLOMICS SOCIETY
From June 19 to 23, the 18th International Conference of the Metabolomics Society was held at the Valencia Conference Center, where leading figures in metabolomics attended.
The Theoreo team, along with Professors Steven Symes and Sean Richards from the University of Tennessee, took part in the prestigious event.
Prof. Jacopo Troisi, CEO of Theoreo Srl, gave a plenary presentation on the latest results related to the MEDEA project highlighting how metabolomics represents a new and promising screening tool for early detection of endometrial cancer.
Prof. Steven Symes outlined the GEMMA project, an international research project with a multi-omics approach to study the onset of autism spectrum disorders.
Prof. Sean Richards presented the results on a validation study, carried out at 15 weeks of pregnancy, to screen for fetal malformations.
Dr. Martina Lombardi, researcher at Theoreo Srl, illustrated an Ensemble Machine Learning model based on metabolomic profiling for noninvasive multicancer screening.

III ANNUAL MEETING OF THE GEMMA PROJECT
The third annual meeting of the GEMMA project, held May 19-20 at the EBRIS Foundation, was a valuable opportunity for interdisciplinary exchange that was attended by all partners afferent to the project.
Theoreo was present with an extensive talk, given by Dr. Martina Lombardi, regarding the perspectives of fecal metabolomics in the study and diagnosis of autism spectrum disorders.
The talk received wide interest, demonstrating the continuing commitment of Theoreo researchers to biomedical study.

STEROID HORMONE-PRODUCING BACTERIA UNDERLYING RESISTANCE TO ANDROGENIC DEPRIVATION THERAPY IN PROSTATIC CARINOMA
Prostate cancer is often treated with ADT, a therapy that negativly affects the biosynthesis of androgens by reducing the possibility of cancer cells growing. Despite the consolidated effectiveness of this treatment, sometimes some subjects, even many years after the first episode, show a resumption of tumor growth.
The study just published in the prestigious journal Science shows that this recovery may be due to an imbalance in the intestinal microbiota that allows some bacterial species to complete the biosynthesis of testosterone starting from endogenous precursors. These results, if further confirmed, could open to new perspectives for the treatment of this fearful pathology and to a new paradigm in the unknown of the complex interaction between human health and the intestinal microbiome.
Theoreo srl proudly participated in this important study coordinated by the Oncological Research Institute based in Lugano in Switzerland, which lasted more than 3 years and which engaged several Italian, Swiss, American and British researchers.

What if SARS-CoV-2 isn't transmitted exclusively by air?
Our researchers, who have always been involved in the study of human health protection strategies, have collected the numerous evidences that would seem to suggest a possible fecal-oral transmission of the virus. This scenario opens up new and interesting perspectives both from a therapeutic point of view and for emergency management with a view to containing infections.
The results of this survey have been published, in collaboration with prof. Alessio Fasano, professor of pediatric gastroenterology at Harvard University, in the "World Journal of Gastroenterolology" and are freely available at the link:

A NEW PATH FOR SCREENING OF BLADDER CANCER
Our researchers, in collaboration with researchers from the University of Tennessee, have taken another small but stubborn step in the fight against cancer by defining a new metabolomic signature for bladder cancer. This opens up new scenarios for understanding the basic mechanisms of neoplastic transformation and for explaining the aggressiveness of some cancers. The results of this study, just published by the journal Applied Science, also indicate an innovative possibility for a population screening system for this disease that affects over 26,000 people a year in Italy alone.

INTESTINAL MICROBIOTA: A STOP TO THE DEVELOPMENT OF COLON-RECTUM CANCER DISCOVERED
Our spin-off partnered with Humanitas University in a study that demonstrated, for the first time, that a strain of gut bacteria plays a protective role against the development of colorectal cancer. The results of the research, supported by AIRC - AIRC Foundation for Cancer Research and directed and coordinated by Prof. Maria Rescigno of Humanitas University, were recently published in the journal "Nature Microbiology".
This study lays the foundations to put a stop to the development of colorectal cancer that affects more than 34,000 people every year in Italy alone.

The European Commission has selected the GEMMA project which aims at identifying potential targets able to favor personalized treatment and prevention of Autism Spectrum Disorders. The study will have a budget of € 14.2M.
GEMMA is an innovative scientific research project for the evaluation of interactions between microbiome, metabolome, epigenome and immune response in order to provide new diagnostic and preventive approaches to autism.

Femal cancers prevention day
